Patch-Clamp Recording and RT-PCR on Single Cells

چکیده
The technique described in this chapter provides electrophysiologists using patch-clamp with a convenient method to link electrophysiological data to a molecular analysis of the mRNAs expressed in a single cell. This molecular analysis can be used either to correlate cell responses with their molecular basis or to identify cell types according to the expression of specific markers. The core of the molecular analysis is polymerase chain reaction (PCR), which makes it fast, sensitive, and simple. Figure 1 outlines the general procedure followed in the experiments. Briefly, after recording of a cell with a patchclamp electrode, the cell content is aspirated through the tip of the electrode and expelled into a test tube with the whole content of the patch electrode. Reagents are then added to perform first strand cDNA synthesis from the mRNA present in the cell. After completion of the reverse transcription (RT) reaction, further reagents are added to the tube to enable a PCR reaction to amplify the cDNA(s) under investigation. Basically, one tube corresponds to one cell, since no change of tubes and very few biochemical manipulations are required. After the first PCR the amplified DNA is analyzed on agarose gel electrophoresis. In some instances, the DNA product from the first PCR is reamplified through a second

Single-cell RT-PCR, a technique to decipher the electrical, anatomical, and genetic determinants of neuronal diversity.
The patch-clamp technique has allowed for detailed studies on the electrical properties of neurons. Dye loading through patch pipettes enabled characterizing the morphological properties of the neurons. In addition, the patch-clamp technique also allows for harvesting mRNA from single cells to study gene expression at the single cell level (known as single-cell RT-PCR). The combination of these...

The real-time quantitative polymerase chain reaction (rtqPCR) has overcome the limitations of conventional, time-consuming quantitative PCR strategies and is maturing into a routine tool to quantify gene expression levels, following reverse transcription (RT) of mRNA into complementary DNA (cDNA). Expression pro®ling with single-cell resolution is highly desirable, in particular for complex tis...

متن کاملDigital PCR to determine the number of transcripts from single neurons after patch-clamp recording.
Whole-cell patch-clamp recording enables detection of electrophysiological signals from single neurons as well as harvesting of perisomatic RNA through the patch pipette for subsequent gene expression analysis. Amplification and profiling of RNA with traditional quantitative real-time PCR (qRT-PCR) do not provide exact quantitation due to experimental variation caused by the limited amount of n...
متن کاملInward rectifying currents stabilize the membrane potential in dendrites of mouse amacrine cells: patch-clamp recordings and single-cell RT-PCR.
PURPOSE To explore the possible existence of inward rectifying currents in the distal dendrites of amacrine cells. METHODS Patch-clamp recordings were made from amacrine cells in a new horizontal slice preparation of mouse retina. Single-cell RT-PCR studies were performed after the patch-clamp recordings. RESULTS In contrast to results from vertical slices or dissociated cells, all amacrine...
